Some of the themes redirect post link to index as a parameter and open content as pop up. The examples I found are very old themes (2011-2012)

Is this “still” acceptable?

For example:

http://themeurl.com/sample-post is redirected to: http://themeurl.com/?post=sample-post or: http://themeurl.com/#sample-post

If it use AJAX, then yes, but the condition is that it should work as expected if the javascript is disabled.

Do I have to be ready for no-javascript-support browsers in all parts of the theme?

Yes. It MUST work without javascript. Javascript should be used only to enchange the user experience.
