I recently got a “rejection” from the reviewers, so I must appeal to other designers eyes for a little help.
What do you guys think I should update to this template design and layout structure:

I like your template but here are a few things I would make changes to:
1. I wouldn’t use the serif font in the navigation. In fact, I might not use the serif font for any of the titles either. It muddies up the clean design.
2. I would center the word “Testimonials” inside of the testimonial bubble instead of having it hang out.
3. The icons in the footer widgets for Twitter, email, and network links I would align left in line with the title instead of way over to the right.
4. I think there’s too much transparency in the navigation dropdown.
Definitely submit it again after making some tweeks especially if this is for the HTML template category.

There is nothing wrong in the typeface. SERIF rules if used in a proper way. Regarding “Testimonials” alignment, that is done deliberately, and looks great. (As an example, check out DDStudio’s latest Fler theme. The gravatars were hanged out of a circle on comments section. That’s art, and looks awesome. There is nothing like “stick to rules of alignment”).
You just need to play a bit with font-sizes, line-height, margin and padding. Otherwise, it looks nice. Especially, jquery effects on navigation. 
By the way, what message you got from reviewer?

I consider “Testimonials” block also looking great. I think it’s out of the box, not like an usual layout. But it seems that this things are not liked but some designers eyes.
I don’t think “You just need to play a bit with font-sizes, line-height, margin and padding…” is enough because I have received the message “rejected” and “need significant improvements”, and not some specific feedback. I supose this means I have to make some major changes to this template.

If you have got that message, then please do not spend more time on this theme. You will only get disappointment. If the item is soft rejected, or rejected because of some minor issues, you can try to correct them. But in cases, when an item is rejected with a message “Needs significant improvements”, you have to work around a lot. That is almost equivalent to starting from scratch.
You can also send a support ticket to the reviewer, asking if any improvements will lead a chance of approval? That way, you will save yourself from spending another two weeks in improvement.

I have decided to give it another try and submit it again.
I will modify the template almost 60-70 % of it, and see what will happen.

I clicked on the link and I really like what I saw there. Its a real unique design with some nice jQuery effects, a cool colour scheme and a modern header.
I wish you good luck with it. I would definitly buy this theme (html).

To bad I had to give up improving that theme, because it was rejected multiple times, with no small suggestions for improving from reviewers.
